Aberrant Neural Connections Linked to <b>Depression</b> | Psych Central <b>...</b>

Aberrant Nerve Connections Linked to Depression Major depressive disorder — also known as clinical depression — is a severe illness that affects an individual in many different ways.

The disorder is characterized by sadness, poor concentration, impaired decision-making and an inability to cope. An individual’s ability to experience pleasurable emotion can be impaired, replaced with prolonged negative thoughts and feelings.

New research, using functional magnetic resonance imaging (fMRI), gives scientists a view of how the brain changes during depression.

The findings, published in the journal Biology of Mood & Anxiety Disorders, show that a depressed brain is characterized by abnormal nerve connectivity.

Researchers from Stanford University compared the fMRI scans of women who were resting (but still awake). Half of the women were diagnosed with depression at the time of the scan and the other group consisted of women who had never had severe depression.

fMRI measures changes in blood flow, and by overlaying images of depressed and unaffected brains, a number of differences came to light.

The images showed that the depressed had decreased connectivity between several key regions of the brain responsible for emotional behavior, learning, memory and decision making.

“In addition to decreased connectivity between emotion processing regions of the brain, we found that depression was linked to an increase in connectivity between the dorsal caudate and an area of the prefrontal cortex,” said lead researcher Daniella Furman, a doctoral student in psychology.

“Deep within the brain, the caudate is thought to be involved in learning, motivation, and emotion while the prefrontal cortex at the front of the head is involved in maintaining goals and likely regulating emotional behavior. Together, these regions may act to filter out irrelevant thoughts or actions.”

Researchers believe greater connectivity may limit the depressed from updating their working memory and, as a result, have sustained negative thoughts.

A Florida State University clinical psychologist has identified factors that could cause some women with post-traumatic stress disorder (PTSD) to have chronic, persistent symptoms while others recover naturally over time.

At the conclusion of a two-year study of women from across the nation, Assistant Professor Jesse R. Cougle found that those with PTSD who reported a history of rape or severe childhood physical abuse were more likely to suffer chronic PTSD symptoms. What's more, women who reported more "re-experiencing" symptoms, such as nightmares and flashbacks, at the initial assessment were more likely to suffer from persistent PTSD symptoms two years after the study began.

"What makes our findings unique is the recovery component," Cougle said. "Most studies of this kind have looked at risk and resiliency or the factors that determine who develops PTSD and who doesn't. We studied factors that influence recovery, or lack thereof, in a sample with PTSD."

"What we found,interestingly, is that more than half of the women in our sample - 58 percent - recovered within two years," Cougle said.

Most people will experience at least one potentially traumatic event during their lifetimes that could result in PTSD. Knowing the factors that predict chronic PTSD will help to identify people who are most in need of assistance and treatment after suffering a traumatic event.

Cougle also found that about half of the women who recovered from PTSD during the study did so without any sort of professional treatment.

"Some women have a natural capacity to recover from PTSD," Cougle said. "They won't require treatment to get better."

Cougle's research, "Factors Associated with Chronicity in Post-Traumatic Stress Disorder: A Prospective Analysis of a National Sample of Women," has been published in the journal Psychological Trauma: Theory, Research, Practice and Policy. He conducted the study with Medical University of South Carolina psychiatry professors Heidi Resnick and Dean G. Kilpatrick.

In addition to re-experiencing symptoms, a PTSD diagnosis includes avoidance and numbing, such as efforts to avoid thoughts, feelings or conversations associated with the trauma; efforts to avoid activities, places or people that arouse recollections of the trauma; an inability to recall an important aspect of the trauma; and feelings of emotional detachment. Additional symptoms include hyperarousal,such as difficulty concentrating; difficulty falling or staying asleep; irritability; and hypervigilance.

A team of researchers from the University of Utah and the University of Massachusetts has identified the first gene associated with frequent herpes-related cold sores.

The findings were published in the Dec. 1, 2011, issue of the Journal of Infectious Diseases.

Herpes simplex labialis (HSL) is an infection caused by herpes simplex virus type 1 (HSV-1) that affects more than 70 percent of the U.S. population. Once HSV-1 has infected the body, it is never removed by the immune system. Instead, it is transported to nerve cell bodies, where it lies dormant until it is reactivated. The most common visible symptom of HSV-1 reactivation is a cold sore on or around the mouth. Although a majority people are infected by HSV-1, the frequency of cold sore outbreaks is extremely variable and the causes of reactivation are uncertain.

"Researchers believe that three factors contribute to HSV-1 reactivation - the virus itself, exposure to environmental factors, and genetic susceptibility," says John D. Kriesel, M.D., research associate professor of infectious diseases at the University of Utah School of Medicine and first author on the study. "The goal of our investigation was to define genes linked to cold sore frequency."

Kriesel and his colleagues previously had identified a region of chromosome 21 containing six genes significantly linked to HSL disease using DNA collected from 43 large families to map the human genome. In the current study, Kriesel and his colleagues performed intensive analysis of this chromosome region using single nucleotide polymorphism (SNP) genotyping, a test which identifies differences in genetic make-up between individuals.

"Using SNP genotyping, we were able to identify 45 DNA sequence variations among 618 study participants, 355 of whom were known to be infected with HSV-1," says Kriesel. "We then used two methods called linkage analysis and transmission disequilibrium testing to determine if there was a genetic association between particular DNA sequence variations and the likelihood of having frequent cold sore outbreaks."

Kriesel and his colleagues discovered that an obscure gene called C21orf91 was associated with susceptibility to HSL. They identified five major variations of C21orf91, two of which seemed to protect against HSV-1 reactivation and two of which seemed to increase the likelihood of having frequent cold sore outbreaks.

"There is no cure for HSV-1 and, at this time, there is no way for us to predict or prevent cold sore outbreaks," says Kriesel. "The C21orf91 gene seems to play a role in cold sore susceptibility, and if this data is confirmed among a larger, unrelated population, this discovery could have important implications for the development of drugs that affect cold sore frequency."

Researchers have identified factors that could cause chronic, persistent symptoms in some women with post-traumatic stress disorder (PTSD) whilst others recover naturally over time. Assistant Professor Jesse R. Cougle, a clinical psychologist from the Florida State University, concluded after a two-year nationwide study of women that those with PTSD who reported being raped or had a severe childhood physical abuse were more likely to suffer chronic PTSD symptoms.

He also discovered that those who reported to "re-live" their experiences as nightmare and flashback symptoms at the initial assessment had a higher tendency to suffer from persistent PTSD symptoms two years after the start of the study.

Apart from re-living symptoms, a PTSD diagnosis also includes several other factors, including evaluating whether the individual tries to avoid thoughts, feelings or conversations linked to the trauma, or appears numb to them, whether they try to avoid activities, places or people that instigate memories of their trauma, or an inability to recall important aspects of the trauma they suffered.

Other symptoms include hyper arousal so that the sufferer experiences difficulty in falling or staying asleep, difficulty in concentrating, hyper vigilance and irritability.

Professor Cougle together with Medical University of South Carolina psychiatry professors Heidi Resnick and Dean G. Kilpatrick's conducted a study published online in the journal Psychological Trauma: Theory, Research, Practice and Policy.

Cougle commented: ??

"What makes our findings unique is the recovery component. Most studies of this kind have looked at risk and resiliency or the factors that determine who develops PTSD and who doesn't. We studied factors that influence recovery, or lack thereof, in a sample with PTSD. What we found, interestingly, is that more than half of the women in our sample - 58 percent - recovered within two years."


?? Most people will experience at least one potentially traumatic event during their lifetime that could lead to PTSD. If the factors that predict chronic PTSD are known, those in need of assistance and treatment can be identified after a traumatic event.

Cougle also observed during his study that approximately half of the women recovered from a PTSD without the aid of any professional.

Cougle said:

"Some women have a natural capacity to recover from PTSD. They won't require treatment to get better."??
